We demonstrated that filamentous actin accumulates in the nuclei of zebrafish early embryos and observed in detail the actin dynamics.The results of live-cell imaging showed that the nuclear actin forms patches in the vicinity of condensed chromosomes after the nuclear envelope breakdown. This study investigated the early kinetics of histone H2AX phosphorylation (γ-H2AX) in response to DNA double-strand breaks from laser microirradiation. The accumulation of γ-H2AX was similar in both ATM-proficient and -deficient cells, indicating that ATM is not the main kinase for γ-H2AX at the early timepoint. However, the presence of the DNA-PK inhibitor caused a delay in the γ-H2AX accumulation, suggesting that DNA-PK plays a rapid role in phosphorylating the histone H2AX at DNA double-strand break sites. Dr. Hayashi's paper has been published in PLOS ONE. A fluorescent chemical dye suitable for the observation of fluorescence in living cells has been identified.
